Carmelo was born about 1784 in Bugnara, L'Aquila, Italy, the son of Isidoro Manna and Carmina Gennaro.
His wife was Giovanna DiTomaso. They were married, but the date and place have not been found. Their seven known children were Carmina Domenica Rosa (1814-?), Carmela Antonia (1815-1841), Rosalba Francesca (1818-1820), Rosalba (1821-?), Isidoro (1823-1826), Maria Domenica (1826-1827) and Isidoro (1831-?).
